General procedure: The mixture of isatin (1 mmol), 3-amino-1-phenyl-1H-pyrazol-5(4H)-one (1 mmol), 1,2-diarylethan-1-one, 2,3-dihydroinden-1-one (1 mmol) or 3,4-dihydronaphthalen-1(2H)-one (1 mmol), H2O (6 mL), HOAc (2 mL) was put in a reaction flask under 90 °C about 5-7 h (monitored by TLC). After completion, the reaction mixture was cooled to room temperature and the products would be isolated out at same time. Then, compound 4 was recrystallized from DMF, however, the pure products of 6 and 8 were filtered from water, dried, without further recrystallization.

The specific steps are:Add 7-chloroindole (54.5 mg, 0.3 mmol) to the pressure tube.Benzamidine hydrochloride (140.9 mg, 0.9 mmol),CuI (5.7 mg, 0.03 mmol)And potassium phosphate (381.6 mg, 1.8 mmol).Add 3mL of DMSO,After magnetic stirring at room temperature for 5 hours,The reaction solution was extracted with dichloromethane.The organic layer was washed with saturated brine.After drying anhydrous sodium sulfate,The solvent is distilled off under reduced pressure to obtain a crude product.The crude product is purified by column chromatography with dichloromethane/petroleum ether=1:20 (V/V) as the eluent to obtain the desired product. The product is a yellow solid.The yield was 68percent.

General procedure: A mixture of isatins 1a-f (1 mmol), active cyanomethanes 2a-b (1 mmol), and enolizable ketones 3a-d (1 mmol) in ethanol (3 ml)was stirred at room temperature in the presence of HIL (Scheme 2).The reaction progress was monitored by thin-layer chromatography (TLC). After completion of the reaction, the reaction mixture was treated with ethanol to recover the insoluble catalyst by filtration.The filtrate was then concentrated under vacuum and the desired product was obtained. The structures of the products were confirmed by 1H NMR, IR spectroscopy, and supported by elemental analysis. The filtered catalyst was dried under vacuum for 2 h and was reused with a fresh charge of solvent and reactants for subsequent runs under the same reaction conditions to observe its catalytic efficiency.

General procedure: To a stirred mixture of isatins (1mmol) and malononitrile/ethyl cyanoacetate (1mmol) in methanol (3ml), was added piperidine (10molpercent) and the reaction mixture was stirred at room temperature for 10min. Then 5,7-dihydroxy-4-methyl-2H-chromen-2-one (1mmol) was added to it. The resulting mixture was stirred at reflux until the reaction was completed as indicated by TLC. The resulting precipitate was treated with dilute HCl and washed with water to afford the pure product for analysis.

General procedure: To a well-stirred solution of isatin and malononitrile(1 mmol each) in ethanol (95percent, 4 mL) was added dimedone,4-hydroxycoumarin, 4-hydroxy-N-methylquinolin-2-one,or 2-methyl-pyrazol-2-one [generated in situ from ethylacetoacetate and hydrazine hydrate, 1 mmol each]. To thissolution was added THAM (30 mol percent) and stirring wascontinued at ambient temperature. Upon completion of thereaction (TLC), water (5 mL) was added and stirring wascontinued for 10 min more. Resultant solid product wasfiltered, washed repeatedly with water, and dried. The dried solid was washed thrice with hexaneechloroformmixture (1:1, v/v) and dried again. Resultant product didnot require any further purification.

The yellow precipitates were collected by filtration and recrystallized from ethanol to give pure 3a-3m.

General procedure: (Table 3, Entry 1): beta-Cyclodextrin (1.0 mmol) was dissolved in water (15 mL) by warming to 50-55 OC until a clear solution was formed. Then, isatin (1.0 mmol) dissolved in methanol (0.5 mL) was added followed by semicarbazide (1.0 mmol) and the mixture was stirred at 50-55 OC until the reaction was complete (as monitored by TLC). After completion of the reaction, mixture was cooled to room temperature and beta-cyclodextrin was filtered. The aqueous layer was extracted with ethyl acetate (3 x 10 ml). The combined organic layers were extracted with water, saturated brine solution and dried over anhydrous Na2SO4. The combined organic layers were evaporated under reduced pressure and the resulting crude product was purified by column chromatography by using ethyl acetate and hexane (4:6) as eluent. The aqueous layer was cooled to 5 OC to recover beta-CD (Table 1). The product was confirmed by IR, 1H 13C NMR, mass spectra.
