To a solution of methyl indoline-5-carboxylate (200 mg, 1.13 mmol) and acetone (0.17 mL, 2.2 6mmol) in dichloromethane (10 mL) was added acetic acid (0.39 mL, 6.77 mmol). The reaction mixture was stirred at room temperature for 30 minutes before cooling to 0 C in an ice-water bath. Sodium cyanoborohydride (213 mg, 3.39 mmol) was added in 3 portions. The reaction mixture was stirred at room temperature overnight. The reaction mixture was diluted with dichloromethane (30 mL), washed with saturated solution of NaHCCh (3 x 30 mL) followed by brine (30 mL) and then dried over anhydrous MgS04. Purification by column chromatography (0-70% ethyl acetate in cyclohexane) gave methyl 1 -isopropylindoline-5-carboxylate (45 mg, 0.21 mmol, 18% yield) as a colourless oil.
